检测原理:本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗小鼠IL-1β抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的IL-1β与固相抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中IL-1β的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570 - 630 nm)测定吸光度值。

IL1B 分子靶点综述
白细胞介素1(IL-1)是17kDa的胞外多肽,分为IL-1α和IL-1β两种蛋白。IL-1β是活化的巨噬细胞分泌的蛋白前体,通过caspase1(CASP1/ICE)水解产生的活化形式。该细胞因子是炎症反应的重要介导者,参与许多细胞活性,包括细胞增殖、分化和凋亡。IL-1是一类重要的蛋白家族,它们由生物活化的单核细胞所衍生,参与炎症反应和免疫应答。IL-1β表达升高会产生各种不同的自身炎症综合征,最显著的是单基因条件称为CAPS,是由于触发IL-1β加工的炎性体受体NLRP3发生突变而引起的。本试剂盒用来检测人IL-1β的成熟形式和未裂解的前体形式。

Located in extracellular space and secretory granule. Is expressed in several structures, including dorsal aorta; early conceptus; hemolymphoid system; integument; and reproductive system. Human ortholog(s) of this gene implicated in several diseases, including autoimmune disease (multiple); blood platelet disease (multiple); eye disease (multiple); gastrointestinal system cancer (multiple); and lung disease (multiple). 本产品可以用来检测小鼠种属的血清样本,对小鼠品系没有特殊要求,此外还可以检测小鼠血浆、细胞上清样本,但需要注意的是正常小鼠血清中 IL-1β蛋白的表达量较低,说明书中的样本上样量只是参考,实际样本间表达有差异,建议先选几个样本做个高中低的浓度梯度,摸索一下您的样本合适的上样量。根据个人经验,一般像 IL-1β这种在正常样本中表达量较低的指标选择高敏试剂盒【EK201BHS】的话,样本的可测率相对而言会更高,但也是需要先做预实验摸索样本合适的上样量的。
