检测原理:本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗小鼠IL-10抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的IL-10与固相抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中IL-10的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570 - 630 nm)测定吸光度值。

IL10 分子靶点综述
白细胞介素10(IL-10)是由2个长178个氨基酸的亚基构成的同源二聚体。在人类中,它由位于1号染色体的IL-10基因编码,由5个外显子组成。IL-10主要由单核细胞分泌,淋巴细胞、II型辅助性T细胞(Th2)、肥大细胞、CD4+CD25+Foxp3+调节性T细胞和部分活化的T细胞和B细胞分泌较少。它对免疫调控和炎症具有多种效应。它下调巨噬细胞的Th1细胞因子、II类MHC抗原和共刺激分子。它还能增强B细胞存活、增殖和抗体分泌。IL-10能阻断NF-κB活性,并参与JAK-STAT信号通路调控。IL-10的免疫抑制特性提示其可能应用于临床,抑制器官移植后的排斥。IL-10还具有强烈的消炎特性。

Acts upstream of or within several processes, including cellular response to hepatocyte growth factor stimulus; negative regulation of cytokine production; and response to bacterium. Located in extracellular space. Is expressed in colon; liver; thymus; and thymus primordium. Used to study inflammatory bowel disease. Human ortholog(s) of this gene implicated in several diseases, including artery disease (multiple); autoimmune disease (multiple); eye disease (multiple); gastrointestinal system cancer (multiple); and hematologic cancer (multiple). Orthologous to human IL10 (interleukin 10).
